Strain Description: This strain carries a chimeric promyelocytic leukemia-retinoic acid receptor alpha transgene (PML-RARa) under the control of sequences from the human cathepsin G gene, which directs expression of the transgene to promyelocytes. The source of the transgene is a human cDNA generated as a result of a specific t(15;17) chromosomal translocation found in most humans cases of acute promyelocytic leukemia (APML). Increased percentages of immature and mature myeloid cells are found in the peripheral blood, bone marrow and spleen. Approximately 15% of the transgenic mice develop acute myeloid leukemia after a long latency (starting around 6 months of age). Splenic promyelocytes and intact mice respond to all-trans retinoic acid (ATRA). The penetrance of APML increases to about 60% when this strain is mated to the strain carrying a bcr-3 isoform of RARa-PML (hCG-RARa-PML).
