Description: |
By homologous recombination a 3.65 kb segment of the Gadd45a gene, including upstream sequences and exons 1-3, were replaced with a PGK-neo cassette. Homozygous Gadd45a-deficient mice were born at Mendelian frequency and grew to adulthood normally. One year after 6-8 week-old mice had been subjected to ionizing radiation, the frquency of thymic lymphomas was 3x higher than in wild-type controls. Cell from Gadd45a-deficient mice show chromosomal instability. Approximately 1/2 of Gadd45a-deficient (homozygous) females in a mixed 129:B6 background did not breed successfully. Successful delivery in most in a pure B6 background.

Husbandry: |
Breeder pairs are supplied as a heterozygote (male or female) and wildtype C57BL/6 mate. Average litter size is six. 50% of homozygous females in mixed 129;B6 background died of delivery failure. This trait appears to not be present in the pure C57BL/6 background. |
